Sophisticated cellular signaling networks are evolved to convert perceived environment signals into proper cellular responses that promote survival and maintain normal functions of the cells and the organisms. The ability to engineer novel cellular signaling not only contributes to the understanding of the signaling mechanisms but also hold great promise for therapeutic applications. We have developed chemical approaches towards this goal that integrate chemical or photochemical reactivity-based signal sensing and chemically induced protein proximity methods. The modular design of our approaches allows the rapid generation of novel signaling circuitry that will give predictable and tailored biological functions in responding to endogenous or exogenous cellular stimuli.
